制法
取桑叶、青蒿各70~100g,加水煎煮,滤过,煎液拌入净大豆1000g中,俟吸尽后,蒸透,取出,稍晾,再置容器内,用煎过的桑叶、青蒿渣覆盖,闷使发酵至黄衣上遍时,取出,除去药渣,洗净,置容器内再闷15~20天,至充分发酵、香气逸出时,取出,略蒸,干燥,即得。
性状
本品呈椭圆形,略扁,长0.6~1cm,直径0.5~0.7cm。表面黑色,皱缩不平,种皮未破损者,可见一侧有长椭圆形种脐。质稍柔软或脆,断面棕黑色。气香,味微甘。
贮藏
置通风干燥处,防蛀。
鉴别
(1)取本品粉末约1g,加70%乙醇10ml,超声处理15分钟,离心,取上清液,作为供试品溶液。另取淡豆豉对照药材1g,同法制成对照药材溶液。再取脯氨酸对照品和缬氨酸对照品,加70%乙醇制成每1ml各含1mg的混合溶液,作为对照品溶液。照薄层色谱法(通则0502)试验,分别吸取上述三种溶液各10μl、10μl、2μl,分别点于同一硅胶G薄层板上,以正丁醇-冰醋酸-水(3∶1∶1)为展开剂,展开,取出,晾干,喷以1%吲哚醌-乙酸(10∶1)的混合溶液,在105℃加热至斑点显色清晰。供试品色谱中,在与淡豆豉对照药材色谱和对照品色谱相应的位置上,显相同颜色的斑点。 (2)取本品粉末约1g,加乙醇25ml,超声处理30分钟,滤过,滤液蒸干,残渣加乙醇1ml使溶解,作为供试品溶液。另取淡豆豉对照药材1g、青蒿对照药材0.2g,同法分别制成对照药材溶液。再取大豆苷元对照品和染料木素对照品,分别加乙醇制成每1ml含0.5mg的溶液,作为对照品溶液。照薄层色谱法(通则0502)试验,吸取上述五种溶液各5~10μl,分别点于同一硅胶GF254薄层板上,以甲苯-甲酸乙酯-甲酸(10∶4∶0.5)为展开剂,展开,取出,晾干,置紫外光灯(365nm)下检视。供试品色谱中,在与青蒿对照药材色谱相应的位置上,显相同颜色的蓝色荧光主斑点;再置紫外光灯(254nm)下检视,供试品色谱中,在与淡豆豉对照药材色谱和对照品色谱相应的位置上,显相同颜色的斑点。
含量测定
照高效液相色谱法(通则0512)测定。 色谱条件与系统适用性试验 以十八烷基硅烷键合硅胶为填充剂;以乙腈-1%冰醋酸(25∶75)为流动相;检测波长为260nm。理论板数按大豆苷元峰和染料木素峰计算均应不低于5000。 对照品溶液的制备 取大豆苷元对照品、染料木素对照品各10mg,精密称定,置50ml量瓶中,加甲醇溶解并稀释至刻度,摇匀,精密量取1ml,置10ml量瓶中,加甲醇至刻度,摇匀,即得(每1ml中含大豆苷元与染料木素各20μg)。 供试品溶液的制备 取本品粉末(过二号筛)约1g,精密称定,置具塞锥形瓶中,精密加入甲醇25ml,称定重量,加热回流1小时,放冷,再称定重量,用甲醇补足减失的重量,摇匀,滤过,取续滤液,即得。 测定法 分别精密吸取对照品溶液与供试品溶液各10μl,注入液相色谱仪,测定,即得。 本品按干燥品计算,含大豆苷元(C15H10O4)和染料木素(C15H10O5)的总量不得少于0.10%。

性状
本品呈椭圆形或类球形,稍扁,长6~12mm,直径5~9mm。表面黑色或灰黑色,光滑或有皱纹,具光泽,一侧有淡黄白色长椭圆形种脐。质坚硬。种皮薄而脆,子叶2,肥厚,黄绿色或淡黄色。气微,味淡,嚼之有豆腥味。
检查
水分 不得过9.0%(通则0832第二法)。 总灰分 不得过7.0%(通则2302)。
贮藏
置通风干燥处,防蛀。
鉴别
(1)本品粉末黄绿色。种皮栅状细胞紫红色,侧面观细胞1列,长50~80μm,壁厚,具光辉带;表面观呈多角形或长多角形,直径约至18μm。种皮支持细胞1列,侧面观呈哑铃状或骨状,长26~185μm;表面观呈类圆形或扁圆形,直径10~28μm,可见两个同心圆圈。子叶细胞含糊粉粒和脂肪油滴。草酸钙结晶,存在于子叶细胞中,呈柱状、双锥形或方形,长3~33μm,直径3~10μm。 (2)取本品粉末2g,加甲醇20ml,超声处理30分钟,滤过,滤液蒸干,残渣加甲醇1ml使溶解,作为供试品溶液。另取黑豆对照药材2g,同法制成对照药材溶液。再取大豆苷对照品、大豆苷元对照品,加甲醇分别制成每1ml各含1mg的溶液,作为对照品溶液。照薄层色谱法(通则0502)试验,吸取上述四种溶液各5μl,分别点于同一硅胶G薄层板上,以甲苯-甲醇-甲酸(14∶6∶0.1)为展开剂,展开,取出,晾干,置紫外光灯(254nm)下检视。供试品色谱中,在与对照药材色谱和对照品色谱相应的位置上,显相同颜色的荧光斑点。
