- Review Question
- What is the effect of agmatine on learning and memory in animals? Context and rationale Agmatine is known as a polyamine derived from decarboxylation of L-arginine. It is widely distributed in the brain with a concentration similar to the other neurotransmitters (Feng, Halaris et al. 1997). Considering that agmatine is produced in the brain, stored in synaptic vesicles, and released by cell depolarization, it might be considered as a supposed neurotransmitter (Reis and Regunathan 2000). It was suggested that agmatine, as an endogenous polyamine, might participate in the processes of learning and memory as a neurotransmitter (Liu, Chary et al. 2008, Liu, Collie et al. 2008) and that aging affects agmatine levels in memory-related structures dramatically in a region-specific manner (Liu, Chary et al. 2008). (Liu, Chary et al. 2008). In spite of the given comprehensive pharmacological effects of agmatine, there are some controversies about the effect of agmatine on memory. Up to now, there have been no systematic reviews to analyze the effectiveness of agmatine on learning and memory function. Reviews based on animal data could make trials’ planning more perfect, increase the odds of success of future clinical trials and assist to decide what is valuable in further research (Murphy and Murphy 2010). Additionally, animal experiment can make us better understand the mechanism of agmatine on learning and memory and guide the future clinical study. Therefore, we are going to conduct a systematic review and meta-analysis of the effectiveness of agmatine on learning and memory in animal experiments to provide suggestions for future animal experiments and clinical trials.

- Inclusion criteria: Between-subject, cross-sectional designs. Exclusion criteria: Within-subject, cross-over designs.

- Inclusion criteria: Animal treated with agmatine (all species, regardless of gender, age and weight) Exclusion criteria: 1. Human studies 2. In vitro experimental studies
- Human Disease Modelled
- Influence of agmatine treatment on memory function
- Intervention
- Inclusion criteria: Treatment with agmatine to experimental animals subjected to memory assessment. All timings, frequencies and dosages of treatment are eligible for inclusion. Exclusion criteria: Co-treatment with agmatine.
- Comparator Control
- Inclusion criteria: Our control group contain non agmatine treated animals. Aim of this study is to include the studies comparing agmatine treated animals to non agmatine treated ones. Exclusion criteria: Absence of Vehicle-treated control animals
- Main Outcome
- Outcome Measure
- Inclusion criteria: All studies assessing the following outcomes : indexes of learning and/or memory function tests, including Morris water maze (MWM), Y maze, step down test, dark avoidance test, active avoidance reaction and one step through test (Object recognition memory task, Elevated plus maze, T-maze task, Radial arm maze Forced swimming test). The secondary outcome measures will be mechanisms of agmatine for learning and/or memory function. Exclusion criteria: If study measured none of above outcomes, it will be excluded.
