- Only randomized controlled trials examine the effectiveness of leucine supplementation compared to a placebo or control group will be included in the review.
- Condition Being Studied
- Leucine has been thought to augment adaptations to strength training by acting as the primary signal to activate protein synthesis (e.g. regulation of translation initiation). Leucine (Leu), have been shown to enhance MPS and reduce muscle protein breakdown by stimulating anabolic factors and reducing catabolic factors. Moreover, leucine appears to decrease skeletal muscle soreness following eccentric exercise, and prevent declines in both circulating testosterone and skeletal muscle power following an overreaching cycle. Leucine which an essential amino acid is one of the most consumed sports supplements among athletes in various endurance and strength disciplines. Hence, the study of ergogenic and physiological effectiveness of leucine is crucial to athletes in different sports.
- Participant
- The review will include studies that examine the acute effects of leucine supplementation during or immediately following strength training activity, involving individuals aged 18 years and older, encompassing sedentary individuals, recreationally active individuals, and athletes, of both male and female genders. Studies involving subjects with recent or ongoing medical issues that might have had a major impact on how they responded to leucine supplementation will be excluded.

- Only randomized controlled trials will be included in the review. Studies included in the analysis must implement leucine supplementation as a distinct intervention. Studies where leucine or its metabolites are administered alongside other supplements or interventions will be excluded to accurately assess the acute effects of leucine supplementation on exercise recovery. Studies looking at the effects of long-term leucine supplementation will not be included.
- Comparator Control
- Leucine may be provided alone or as its metabolites, to the selected participant population. The specific leucine metabolites considered may include α-ketoisocaproic acid (KIC) or β-hydroxy-β-methylbutyrate (HMB), among others, as applicable. Studies included in the analysis must implement leucine supplementation as a distinct intervention. Studies where leucine or its metabolites are administered alongside other supplements or interventions are excluded to accurately assess the acute effects of leucine supplementation on exercise recovery. The review will include studies comparing the use of leucine supplements to placebos or control groups.
- Main Outcome
- Studies examining the acute effects of leucine supplementation during or right after strength training activity will be considered within the scope of the review. The following are the key outcome measurements pre-planned for the meta-analysis; -Creatine kinase (CK) -MVC -Soreness Measures of effect To determine the effect of acute effects of leucine supplementation intake during or right after strength training activity on pain, strength, and creatine kinase (CK) results. For creatine kinase values, the effect measure can be the mean difference (MD) or standardized mean difference (SMD) between leucine and placebo or control groups. For MVC values, the effect measure can be MD or SMD between leucine and placebo or control groups. For soroness values, the effect measure can be MD or SMD between leucine and placebo or control groups.
