- Title
- Monitoring of the efficacy of artemisinin-based combination therapies and trend of resistance molecular markers of Plasmodium falciparum to antimalarial drugs over ten years in sub-Saharan Africa: a systematic review and meta-analysis
- Review Question
- What are the efficacy and evolution of resistance molecular markers of malaria drugs in sub-Saharan Africa over ten years?
- Study Type Included
- Inclusion criteria For efficacy and effectiveness studies o Original articles of studies that investigated the efficacy or effectiveness of at least one recommended ACT for the treatment of uncomplicated P. falciparum; o Study samples collected between 2010 – 2021 o Studies performed in Sub-Saharan Africa For Resistance molecular markers review o Clinical trials comprising analysis of one or more malaria resistance molecular markers (Pfcrt, Pfmdr, Pfdhps, Pfdhfr, Pfk13, Pfpm2) o Observational studies (Cross-sectional/cohort studies/) involving detection of the prevalence of the mutation in at least one of the genes (Pfcrt, Pfcrt, Pfmdr, Pfdhps, Pfdhfr, Pfk13, Pfpm2)
- Condition Being Studied
- The systematic review focuses on malaria treatment. It contains two parts. First, we will analyse the efficacy of artemisinin-based combination therapy five years after its scale-up in many countries. Second, we will examine the trend of molecular resistance markers over ten years using ACT.
- Participant
- - Inclusion: patients with Plasmodium falciparum infection, regardless of age or sex, living in sub-Saharan Africa - Exclusion: Expats living in Africa
- Animal
- Human Disease Modelled
- Intervention
- Use of artemisinin agents in the treatment of uncomplicated malaria
- Comparator Control
- Main Outcome
- - Efficacy review Uncorrected and corrected Adequate clinical and parasitological response (ACPR) by Day 28 or 42 - Resistance molecular markers review Prevalence of malaria resistance molecular markers (Pfcrt, Pfmdr1, PfK13, dhps, dhfr, plasmepsin 2, Pfmdr1 number of copy) Measures of effect - Proportion of ACPR - Prevalence
- Outcome Measure

- Funding Source
- PAMAfrica/EDCTP This systematic review is funded through the EDCTP-funded project PAMAfrica (Portofolio approach to developing the next generations of malaria treatments for Africa). This study is part 1 of the PhD project, which aims at assessing the temporal trends of Plasmodium falciparum resistance molecular markers over ten-year sin Burkina Faso.
