Title
The Efficacy of Alpha Lipoic Acid in Nonalcoholic Liver Disease and Nonalcoholic Steatohepatitis: A Systematic Review and Meta Analysis of Clinical Trials
Review Question
What is the current state of evidence regarding alpha lipoic acid as a treatment for nonalcoholic fatty liver disease/metabolic dysfunction-associated steatotic liver disease and nonalcoholic steatohepatitis?
Study Type Included
Clinical trials
Condition Being Studied
Non-alcoholic fatty liver disease (NAFLD) is the most common liver disease with a prevalence of 25% and characterized by different stages of progression such as non-alcoholic fatty liver (NAFL) (steatosis) and nonalcoholic steatohepatitis (NASH), that leads to cirrhosis. NAFLD and NASH are related to insulin resistance and metabolic syndrome. Some animal studies suggested that alpha-lipoic acid (ALA) can improve these disease outcomes. In addition, there is evidence of beneficial effects of ALA on metabolic disorders and insulin resistances.
Participant
Inclusion criteria: clinical trials, NAFLD/NASH patients Exclusion criteria: irretrievable full-text articles, incompatible language
Animal
Human Disease Modelled
Intervention
The interventions are products that consist of alpha lipoic acids.
Comparator Control
Placebo or other medications
Main Outcome
Changes from baseline in metabolic parameters (e.g. lipid profile, HbA1c, glucose parameter) and liver functions. Measures of effect No specific timing constrained on the duration of the intervention course. Effect sizes will be presented in mean differences or standardized mean differences.
Outcome Measure
Additional Outcome
Data permitting, changes from baseline in: Anthropometric parameters (e.g. Body weight, and BMI) Markers of oxidative stress (e.g. malondialdehyde (MDA)) Inflammatory markers (e.g. C-reactive protein (CRP), Interleukin-1, Interleukin-6, monocyte chemoattractant protein-1 (MCP-1)) Iron profile (e.g. serum ferritin) Hematological profile (e.g. platelets) Radiological modalities (e.g steatosis score) NAFLD scoring tools (e.g NAFLD fibrosis score) Liver biopsy changes Measures of effect No specific timing constrained on the duration of the intervention course. Effect sizes will be presented in mean differences or standardized mean differences.
