Efficacy and safety of caffeic acid tablets in the treatment of leukopenia: a systematic review and meta-analysis
Review Question
P:The patients diagnosed with leukopenia (leukocyte count < 4.0×109/L) were included in the study regardless of age and gender. I:Intervention measures: Patients in the test group were treated with caffeic acid tablets (200-300 mg/time, 3 times/d. Dezhou Deyao Pharmaceutical Co., Ltd., approved by Chinese medicine H37020537, production batch number: 2102140503.) or caffeic acid tablets combined with conventional drugs for leukopenia. C:conventional drugs for leukopenia. The dose and duration of administration were not limited. O:The main efficacy indicators were clinical efficiency rate and changes in white blood cell count, and secondary efficacy indicators were the incidence of adverse reactions, grade III and IV myelosuppression, incidence of infection, platelet count, and neutrophil count.

Condition Being Studied
The main ingredient of caffeic acid tablet is caffeic acid, also known as "3, 4-hydroxycinnamic acid" or "3, 4-dihydroxy phenyl acrylic acid", which is a phenolic compound and classified as cinnamic acid . Its molecular formula: C9H8O4, relative molecular weight: 180.16Da. It is widely found in many plant foods and herbs such as fruits, wine, coffee, olive oil, and legumes . Caffeic acid has antidepressant, antihyperglycemic, antioxidant, anticancer, antiviral, anti-inflammatory, and immunomodulatory properties. The efficacy and safety of caffeic acid tablets in the treatment of leukopenia have widely been reported in the literature, but no systematic evaluation is available. Therefore, in this study, a meta-analysis was conducted to compare the efficacy and safety of caffeic acid tablets with other medicines for leukopenia, to provide a reliable basis and evidence-based guidelines for clinical use.
Participant
The patients diagnosed with leukopenia (leukocyte count < 4.0×109/L) were included in the study regardless of age and gender. According to the leukocyte grading criteria set by the World Health Organization, the leukocyte counts were graded as follows: degree 0: > 4.0×109/L; degree I: 3.0 - 3.9×109/L; degree II: 2.0 - 2.9×109/L; degree III: 1.0 - 1.9×109/L; degree IV: < 1.0×109/L

Comparator Control
the control group was treated with conventional drugs for leukopenia.
Main Outcome
(I)The main efficacy indicators were clinical efficiency rate and changes in white blood cell count, and secondary efficacy indicators were the incidence of adverse reactions, grade III and IV myelosuppression, incidence of infection, platelet count, and neutrophil count. The clinical efficiency was indicated in the following manner: (i) significantly effective if the total leukocyte count has increased > 1.0×109/L and lasted for 2 weeks; (ii) effective if the total leukocyte count has increased between 0.5 - 1.0×109/L and lasted for 2 weeks; (iii) ineffective if the total leukocyte count has increased < 0.5×109/L. Measures of effect Meta-analysis was performed using RevMan 5.3 and Stata 17.0 software. Count data were analyzed by relative risk ratio (RR), and continuous data were analyzed by mean difference (MD) or standardized mean difference (SMD). Also, their combined effect sizes and their 95% confidence intervals (CI) were calculated. Heterogeneity was analyzed using the Q test and I² test.
