- Review Question
- In patients with osteoarthritis does intra-articular triamcinolone extended release (ER) control pain and improve function for a longer duration than standard triamcinolone? Is triamcinolone ER safer alternative to standard triamcinolone for use in patient's with typical contraindications (ex, DM, uncontrolled HTN)? Does Triamcinolone ER decrease the common side effect risk of hyperglycemia in patients with DM compared to standard Triamcinolone? Does Triamcinolone ER decrease the potential side effect of cartilage damage in comparison to standard triamcinolone?
- Study Type Included
- We will include randomized trials to assess the role, efficacy, and safety of intra-articular Triamcinolone Acetonide Extended Release (TA-ER) and will supplement these with observational studies (including cohort studies).
- Condition Being Studied
- Osteoarthritis (OA) is a degenerative joint disease associated with pain, disability, and loss of function that represents a significant and increasing public health burden. Osteoarthritis can affect any joint with knee the most common joint affected followed by hip, and hand respectively.

- - Adults age 30-75 males and females with Osteoarthritis defined by the American College of Rheumatology clinical classification criteria for OA - Adults with osteoarthritis plus T2DM and/or HTN

- Intervention
- Intervention: Extended Release Triamcinolone (TA-ER) Inclusion Criteria: - Adults age 35-75 males and females with Osteoarthritis defined by the American College of Rheumatology clinical classification criteria for OA - Adults with Osteoarthritis, T2DM and/or HTN Exclusion Criteria: - History of chronic inflammatory disease - Pregnant or breastfeeding women - Patients with intra-articular injection within the last 3 months
- Comparator Control
- Control: Triamcinolone Acetonide Crystalline Suspension (TAcs), Placebo (intra-articular saline)
- Main Outcome
- Differences in pain, stiffness, and physical function measured using WOMAC scores. Differences in osteoarthritis Outcome Score-assessed Quality of Life (KOOS-QOL) scores. Differences in blood glucose levels using glucose monitor values and/or serum testing. Safety/Adverse events with TA-ER compared to TAcs. Measures of effect We will measure outcome using relative risks, number needed to treat, and odds ratios.
