Title
The effects of changes in the concentration of tryptophan derivatives on chronic musculoskeletal pain – a systematic review and meta-analysis
Review Question
P= patients, I = Investigated condition, C= control, O =outcome In adult individuals (P) with chronic musculoskeletal pain (I), do levels of tryptophan derivatives (O) differ when compared to healthy pain-free individuals (C)
Study Type Included
Inclusion criteria · Observational, cross-sectional, and case-control studies as well as clinical trials (prospective, retrospective and randomized controlled trials (RCT)). · Studies including measurement of the levels of tryptophan derivative levels. Exclusion criteria · Studies presented in other languages than English and Scandinavian languages · Editorials, letters, legal cases, interviews, case-series, duplicates
Condition Being Studied
Chronic pain is a prevalent affliction that greatly impacts people's daily lives and can, in some instances, even lead to depression. Despite it being such a common issue, most patients do not get the appropriate treatment. Therefore, a deeper knowledge of chronic pain and its associations is crucial for new methods on how to diagnose, manage, and treat chronic pain. Tryptophan is one of our bodies' nine essential amino acids to function properly. Tryptophan helps the body produce metabolites such as melatonin, kynurenine, and serotonin, and its deficiency can have many effects on the body such as depression, anxiety, and the inability to concentrate. Chronic pain has similar pathophysiological patterns to depression, and the activity of the tryptophan metabolites has been shown to strongly influence comorbidities such as depression and chronic pain. A broader understanding of the correlation between tryptophan metabolites and chronic musculoskeletal pain would allow us to utilize new diagnostic techniques which are required to further grasp the disorders that affect our bodies and how to treat them. This study will focus on how changes in the concentration of tryptophan derivatives can affect chronic musculoskeletal pain in patients through a systematic review and meta-analysis.
Participant
Inclusion criteria are adult (≥ 18 years old) individuals of both sexes with chronic musculoskeletal pain.

Human Disease Modelled
Intervention
Concentration of tryptophan derivatives.
Comparator Control
Inclusion criterion is pain-free individuals. Exclusion criterion is any kind of painful condition.
Main Outcome
The main outcome is reported level on one occasion of tryptophan derivatives in chronic musculoskeletal pain patients and controls. Measures of effect The relative risk based on the quote between levels in musculoskeletal pain patients vs. controls.
