- Trimethylamine-N-oxide (TMAO) and multiple health outcomes: umbrella review of meta-analyses of observational studies and Mendelian randomization studies
- Review Question
- The primary objective of this systematic review is to summarize the evidence and evaluate the validity of the associations between TMAO and multiple health outcomes. Furthermore, we will perform update meta-analyses to assess the association between TMAO concentrations and multiple health outcomes.
- Study Type Included
- Inclusion (umbrella review): meta-analyses. We will include one type of study in the umbrella review: meta-analyses exploring the associations between trimethylamine N-oxide (TMAO) concentrations and any health outcomes in human participants. Exclusion (umbrella review): conference abstracts, interviews, letters, reviews. Inclusion (meta-analyses): case-control (including case-cohort and nested case-control) studies, cohort studies. We include one type of study to perform the updated meta-analyses: observational studies exploring the associations between TMAO concentrations and any health outcomes. Exclusion (meta-analyses): conference abstracts, interviews, letters, reviews and meta-analyses. We will exclude studies that do not present study specific data (relative risks, odds ratios, hazard ratios or 95% confidence intervals).

- Multiple health-related outcomes: mortality, cardiovascular outcomes (cardiovascular disease, hypertension, stroke, major adverse cardiovascular events and so on), anthropometric variables (blood pressure, body mass index), metabolites and metabolic diseases (lipoprotein cholesterol, triglycerides, c-reactive protein, diabetes mellitus and so on), renal outcomes.

- Intervention
- Intervention: trimethylamine oxide, trimethylamine N-oxide, trimethylammonium oxide and TMAO.
- Comparator Control
- The comparison is the concentrations of TMAO in cases and their controls.
- Main Outcome
- Incidence of stroke, hypertension, cardiovascular disease, major adverse cardiovascular events, diabetes mellitus, and mortality, blood pressure, body mass index, low/high- density lipoprotein cholesterol, total cholesterol, triglycerides, c-reactive protein, and glomerular filtration rate. Measures of effect Measures of categorical variable relative risks, odds ratios, hazard ratios. Measures of continuous variable: weighed mean difference.
