Effects and potential mechanism(s) of ligustrazine for the treatment of diabetic nephropathy: A Preclinical Systematic Review and Meta-analysis
Review Question
Is ligustrazine effective for diabetic nephropathy rats? What are the possible mechanisms of ligustrazine on diabetic nephropathy? Context and rationale Diabetic nephropathy is one of the most common complications of diabetes. The condition of diabetic nephropathy patients often deteriorates with the increase of age and the prolongation of the course of disease. Due to the lack of effective treatment, diabetic nephropathy patients often end up with renal failure. Ligustrazine is the extract of Chinese herbal medicine Chuan-Xiong, which is used in the treatment of myocardial ischemia, diabetic nephropathy and other diseases. Animal researches show that ligustrazine preparation is effective for diabetic nephropathy. However, the targets and mechanisms of ligustrazine in the treatment of diabetic nephropathy have not been fully revealed. In this study, we will systematically collect the experimental evidence of ligustrazine in the treatment of diabetic nephropathy, in order to comprehensively evaluate the targets and mechanisms of its treatment of diabetic nephropathy. This review can provide empirical evidence to improve the likelihood of success of future clinical trials.

Inclusion criteria: Prospective randomized controlled studies. Exclusion criteria: Case reports; Cross-over studies; studies without a separate control group.

Inclusion criteria: All studies about diabetic nephropathy in rats induced by STZ can be included. Exclusion criteria: In vitro studies; Studies in humans; In sillico studies, models of diseases other than diabetic nephropathy induced by STZ.
Human Disease Modelled
This is an animal disease model. diabetic nephropathy.
Intervention
Inclusion criteria: Ligustrazine is the extract of Chinese herbal medicine chuanxiong which is widely used in diabetic nephropathy. Animal studies on the treatment of diabetic nephropathy with ligustrazine will be included. There will no restrictions for the frequency and dosage of ligustrazine. Exclusion criteria: Studies that use con-interventions where the effect of ligustrazine will not be discerned.
Comparator Control
Inclusion criteria: Purified water-treated, saline-treated or no treatment. Exclusion criteria: There is no limitation about treatment in control group.
Main Outcome
Outcome Measure
Inclusion criteria: Body weight, blood glucose, HbA1c, blood urea nitrogen, scr serum creatinine, kidney tubules injury index; relative area of kidney collage; MDA; SOD, eNOS. Exclusion criteria: All other outcomes that are not directly related to diabetic nephropathy.
