- Review Question
- The aim of this study was to conduct a meta-analysis of the efficacy and safety of adjunctive pregnenolone for schizophrenia. Clinical efficacy indicators (PANSS) were used to determine whether the mental symptoms of schizophrenia patients improved, so as to infer whether pregnenolone is an effective, safe and reasonable choice for adjuvant treatment of schizophrenia.

- Condition Being Studied
- As a chronic psychiatric disorder, schizophrenia affects the worldwide population with a lifetime prevalence of 1% and considerable long-term morbidity, mortality, and burden. Patients with schizophrenia often have positive or negative symptoms. Routine treatment regimens are accompanied by adverse effects, and a significant portion of patients remain symptomatic despite treatment, especially those with negative symptoms, which are frequently severe and strongly correlated with decreased functional outcome and quality of life. Despite the large number of psychotropic medications currently available, effective management of schizophrenia continues to be a challenging task. New treatment strategies are therefore necessary to address these important dimensions of schizophrenia symptomatology.

- Participants: patients that suffer from schizophrenia, chronic schizophrenia or schizoaffective disorder.

- Intervention: study group given pregnenolone.
- Comparator Control
- Comparison: control group were given placebo.
- Main Outcome
- The main outcome is the negative symptoms of schizophrenia. Measures of effect The primary rating instrument are PANSS negative subscal scores and SANS score.

- Additional Outcome
- The additional outcomes include the severity of psychiatric symptoms, function, the positive symptoms and general psychopathological symptoms of schizophrenia. Measures of effect The rating instruments are the PANSS positive subscal scores, the PANSS total score, the PANSS general score, and the GAF score.
