Antitumor effects of oleandrin on different types of cancers: Systematic review of experimental animal models and cell cultures
Review Question
What are the effects of oleandrin on different types of cancer in vivo and in vitro models? Is oleandrine effective in reducing tumor size and cell proliferation in different types of cancer? Context and rationale Nerium oleander, a member of the family Apocynaceae, is commonly known as Kaner in various countries of Asia and the Mediterranean region. This plant has been renowned to possess significant therapeutic potential due to its various bioactive compounds which have been isolated from this plant e.g., cardiac glycosides, oleandrin, a-tocopherol, digitoxingenin, ursolic acid, quercetin, odorosides, and adigoside. Oleandrin, a saponin glycoside is one of the most potent and pharmacologically active phytochemicals of N. oleander. Its remarkable pharmacotherapeutic potential has been interpreted as anticancer, anti-inflammatory, anti-HIV, neuroprotective, antimicrobial, and antioxidant. This particular bioactive entity is known to target the multiple deregulated signaling cascades of cancer such as NF-kappa B, MAPK, and PI3K / Akt. The focus of this study is to gather evidence about the effects of oleandrine on different types of cancer in vitro studies in human and animal cells and in experimental studies in animals. The results of in vitro studies and animal studies that tested the effects of oleandrin on cancer are relevant to testing new drugs for treating cancer in humans.

Inclusion criteria: In vitro assays (human cancer cells and animals). In vivo experimental animal models. Exclusion criteria: Clinical trials with humans with cancer.

Inclusion criteria: I - Studies that investigated the anti-tumor effects of oleandrin on breast cancer using in animal models (any animal or mouse or rat) II- In vitro models (human and animal cells) Exclusion criteria: I- Animal: None II- In vitro models that do not use breast cancer
Human Disease Modelled
Cancer occurs when changes called mutations take place in genes that regulate cell growth. The mutations let the cells divide and multiply in an uncontrolled way. It is now well known that cancer is caused by a set of molecular genetic changes that lead to loss of growth control and cellular differentiation, resulting in uncontrollable cell growth that eventually leads to tumor formation.
Intervention
Inclusion criteria: The use of oleandrin in any dosage, timing, frequency will be considered intervention. Exclusion criteria: Oleandrin in combination with other drugs. Studies that did not investigate the antitumor effects of oleandrin on cancer were also excluded.
Comparator Control
Inclusion criteria: Control group or comparison with no treatment, treatment with other drugs and / or treatments known for cancer with radiotherapy or chemotherapy. Exclusion criteria: None
Main Outcome
Outcome Measure
Inclusion criteria: (i) in vitro cell proliferation, cell viability, or tumor volume in vivo in an animal model and (ii) apoptosis and/or cell cycle arrest, including analysis of protein expression and experimental models after treatment with oleandrine. Cellular markers, cell proliferation/apoptosis, and tumor sizes (in vivo) will be collected from the studies. Exclusion criteria: Outcomes other than oleandrin Anti-tumor effects

Other Selection Criteria
Inclusion criteria were: 1) studies that investigated the effects of oleandrin on different types of cancer using in vitro models and animal models; 2) original peer-reviewed research articles; 3) no language restriction; 4) no restriction on the year of publication. In addition, all studies that investigated the effects of oleandrin on cancer were included, regardless of dosage and duration of intervention. For in vitro studies, human and animal cancer cell lines will be considered. The exclusion criteria for the studies were: clinical trials performed in humans and epidemiological studies, theses, dissertations, case studies, editorials, letters to the editor, duplicate studies found in more than one database, non-laboratory studies, articles with the version complete unavailable and withdrawn articles. Studies that used oleandrin in combination with other drugs were also excluded. Studies that did not investigate the antitumor effects of oleandrin on cancer were also excluded.
