- Review Question
- The objective of this review is to synthesise the evidence related to the potential risks involved with ß-alanine supplementation, within the context of its use as an ergogenic aid.
- Study Type Included
- Only intervention based trials (acute and chronic) will be considered for inclusion in this review.
- Condition Being Studied
- Carnosine is a di-peptide comprising ß-alanine and L-histidine, which is primarily present in human skeletal muscle and is thought to exert a pleiotropic effect on excitable tissue metabolism, with purported roles including intracellular buffering, reduction of oxidised particles, and/or calcium regulation. ß-alanine availability is the limiting step in intra-muscular carnosine synthesis. Accordingly supplementation with this amino acid is ever-increasing in popularity and has been reported to convey both ergogenic and therapeutic benefits in a range of populations. Despite the ever-increasing popularity of this nutritional supplement, a systematic risk assessment has yet to be undertaken. The aim of this review therefore is to synthesise the evidence related to the potential risks involved in ß-alanine supplementation, within the context of its use as an ergogenic aid.
- Participant
- Inclusion Criteria: Healthy men and women, of any age or activity level. Healthy animal models. Exclusion Criteria: Studies conducted on clinical populations, or diseased or genetically modified animal models.

- Intervention
- All human and animal trials which investigate the effects of ß-alanine supplementation (both acute and chronic) will be considered for inclusion within this review. Studies investigating multi-ingredient supplements will be excluded, due to the difficulty in isolating outcomes to the effects of ß-alanine. Only studies investigating the effects of oral supplementation strategies will be included.
- Comparator Control
- All intervention trials (both controlled and uncontrolled) will be considered for inclusion within this review, however the quality of evidence for specific studies will be downgraded if they do not employ a blinded, placebo controlled design, as described within the risk of bias section.
- Main Outcome
- Outcome One: Side effects experienced following acute supplementation. Outcome Two: Side effects experienced following chronic supplementation. Outcome Three: Influence of supplementation on related intramuscular amino acid content (mainly taurine and histidine). Outcome Four: Influence of supplementation on health-related biomarkers.

- Additional Outcome
- Data from supplementation studies using animal models will be summarised, however this information is considered to constitute a secondary outcome, due to uncertainty in extrapolating animal data to human physiology.
