Review Question
What are the effects of Lannea Coromandelica on pancreatic islets and ductal cells in Wistar albino rat for diabetes Mellitus? Context and rationale According to WHO, recently compiled data show that approximately 150 million people have diabetes mellitus worldwide, and that this number may well double by the year 2025.In diabetes mellitus there is either impaired synthesis and secretion of insulin (type 1 diabetes mellitus) or impaired sensitivity of tissues to insulin action (type 2 diabetes mellitus), leading to severe metabolic derangement.Despite of ensuing new drug development, advance in technologies and new treatment, diabetes and its associated complications are the major issues even today the developed and developing countries encounter. This metabolic disorder cannot be cured with modern allopathic medicine as the drugs have side effects in long term and do not restore normal level of glucose.To conflict this disorder, natural bio-active small molecules extracted from the herbal plants plays an important role in diabetic therapy. In modern medicine, even traditional herbal plants were known to have valuable therapeutic effects by maintaining better glucose control with low side effects. One such traditional herbal plant is Lannea Coromandelica.It has been reported that Lannea Coromandelica to have potential as wound healing, anti-inflammatory, anti-microbial, hypotensive, ulcerative stomatitis, gout, cholera, diarrhea, sore eyes, leprosy, sprains and elephantiasis. Very few studies have been reported on the anti-diabetic properties of Lannea Coromandelica extract on pancreatic islets and ductal cells. However, earlier studies revealed phytochemical evaluation, pharmacological properties so far.Thus, the present study is designed to substantiate the efficacy of Lannea Coromandelica extract by histo pathological and morphometric analysis.

Inclusion criteria: Experimental Randomized Controlled Trial with Standard Reference drug group. Exclusion criteria: Non-randomized controlled trial

Inclusion criteria: Gender - Adult Male Species -Rat (Rattus norvegicus) Strain - Wistar albino Exclusion criteria: Adult female Species involving other than Wistar albino rats Pups Rats in gestation stage Human participants will be excluded.

Intervention
Inclusion criteria: 1. <u>Experimental induction of diabetes</u>: - Intraperitoneal injection of Streptozotocin (45-65 mg/kg) in 0.1 M cold citrate buffer (pH 4.5) - 5% glucose solution will be given orally for 2 days to prevent hypoglycemia during first 24 hr after Streptozotocin induction. - After 72 hrs, rats with blood glucose level &gt;250 mg/dl will be considered to be diabetic and further will be used in the experiment. 2. <u>Lannea Coromandelica (L.C)</u> - L.C Extract (100-500 mg/kg) will be given by oral gavage Exclusion criteria: Non Standardized agents for inducing diabetes in animal model will be excluded.
Comparator Control
Inclusion criteria: Standard reference drug - Glibenclamide (10 mg/kg) Reference drug group will be included to compare with the experimental groups Exclusion criteria: Vehicle-treated Unhealthy animals Met formin Insulin will be excluded.
Main Outcome
Outcome Measure
Inclusion criteria: 1. <u>Hematological</u> - Blood glucose levels and body weight will be measured on days ( 7, 14, 21 and 28) of the study. - Estimation of glucose levels will be done by using glucometer and glucose oxidase- peroxidase reactive strips. 2. <u>Serum</u> - Insulin, Albumin, Creatinine, TC, HDL, LDL and Triglycerides 3. Behavioral changes will be assessed by Maze test (open, Elevated) 4. Body weight will be measured by using weighing scale. 5. Histological (paraffin blocks - H &amp; E) routine stain 6. Lactate dehydrogenase (LDH) level. 7. Superoxide dismutase Exclusion criteria: Urine analysis Biopsy Globulin Pancreatic enzymes
