- Review Question
- Does Icariside II (ICS II) have a therapeutic effect in animals with acute ischemic stroke? Context and rationale Phosphodiesterase 5 inhibitors (PDE5i) have recently been identified as a new strategy for the treatment of acute ischemic stroke (AIS). Icariside II (ICS II), a novel PDE5i with fewer side effects than conventional PDE5i, has great potential for the treatment of AIS. However, Icariside II (ICS II) has not been used clinically, and pharmacological experiments in animal models of acute ischemic stroke have helped to investigate the efficacy of ICS II.

- Inclusion criteria: The inclusion criteria for the screened publications were: induced ischemic stroke, in vivo animal studies. Exclusion criteria: Exclusion criteria applied were: human studies, in vitro or in situ studies, no induced ischemic stroke, animal models of cerebral hemorrhage.

- Inclusion criteria: Icariside II as an intervention drug, and the single dose should not exceed 40mg/kg and should not be administered more than twice a day. Exclusion criteria: Icariside II (ICS II) was not used as an intervention drug.
- Comparator Control
- Inclusion criteria: Inclusion criteria: The sham-operated group, the model group (MACO), and the drug-administered modeling group, which were treated by gavage, while the sham-operated group rats and model rats were given volume-matched normal saline. Exclusion criteria: Exclusion criteria: saline was not administered in the model and sham-operated groups.

- Inclusion criteria: Outcomes were noted in the following categories: neurogenesis/neuronal protection, functional outcomes (e.g., memory performance). Stroke lesion volume/neurodegeneration, angiogenesis/cerebral vascular protection, cerebral blood flow, apoptosis. Oxidative stress, inflammation within brain tissue. Exclusion criteria: No relevant outcomes.
