- Intraocular Pressure Changes Following Glucosamine Use: A Meta-Analysis of Pre-Post and Placebo-Controlled Studies
- Review Question
- 1- Does glucosamine result in clinically significant changes (defined as 2mmHg change) in the intraocular pressure following its administration as compared to pre-intervention values? 2- Does glucosamine result in clinically significant changes (defined as 2mmHg change) in the intraocular pressure as compared to placebo? 3- Are glucosamine-mediated intraocular pressure changes maintained across different assessment timepoints (short-term or long-term)? 4- Does glucosamine result in any ophthalmic complications or adverse events?

- Both experimental (clinical trials: randomized and non-randomized) and observational (cohort, case series, case-control, cross-sectional) studies.
- Condition Being Studied
- The use of glucosamine has increased in recent decades. Healthcare providers have been recommending it in different disorders, mainly osteoarthritis. Other conditions include rheumatoid arthritis, fibromyalgia, and multiple sclerosis. Although it has shown beneficial effects on joint pain and limb/joint mobility, an increasing number of studies have highlighted a negative effect on the intraocular pressure of affected patients. To date, there is no clear evidence whether or not the use of glucosamine is associated with clinically significant intraocular pressure changes either following it use or when comparing it to placebo. Therefore, we aim to conduct this systematic review and meta-analysis to determine if the use of glucosamine in such patients results in changes in their intraocular pressure either compared with pre-intervention values or with placebo.

- Patients with any condition that requires the use of glucosamine (mainly osteoarthritis)

- Glucosamine (regardless of the dose)
- Comparator Control
- Two comparisons will be included: 1- Pre-intervention vs. post-intervention (glucosamine) 2- Glucosamine vs. placebo (post-intervention)
- Main Outcome
- 1- The intraocular pressure change in pre- vs. post-glucosamine use. 2- The intraocular pressure change between glucosamine and placebo (post-intervention). Measures of effect The data will be reported as mean difference (MD) and its corresponding 95% confidence interval (CI). In instances where intraocular pressure is categorized, the log odds ratio (logOR) and its corresponding 95% CI will be calculated and then subgrouped for each intraocular pressure category.

- Secondary outcomes will include the following: 1- subgroup analysis based on assessment timepoints. 2- subgroup analysis based on study design. 3- eye-related adverse events. Measures of effect In the subgroup analyses, data will be presented as MD and its corresponding 95% CI in each subgroup. As for the adverse events outcome, data will be reported as logOR and its corresponding 95% CI.
