- Review Question
- Xianling Gubao Capsule has been widely used as adjunctive therapy in the treatment of Knee Osteoarthritis (KOA) during the past decades in China, but its clinical efficacy and safety is not yet well confirmed. This study aims to evaluate the efficacy and safety of Xianling Gubao Capsule Combined With Glucosamine in the treatment of KOA.

- Knee osteoarthritis (KOA) is a chronic arthritic disease usually characterized by bone regeneration at the joint margins and subchondral bone as well as articular cartilage fibrosis, ulceration, loss, chafing, and cartilage degeneration. As the global aging process increases, the social problems of aging caused by KOA are becoming more and more evident.
- Participant
- Participants with primary KOA who are diagnosed by clinical guidelines whether traditional Chinese medicine or western medicine will be selected. No age, gender, or ethnicity exemption for participants. Patients will be excluded if they have secondary KOA or other non-suppurative arthritis, such as traumatic arthritis, rheumatoid arthritis, and allergic arthritis.

- The examined treatment intervention was Xianling Gubao Capsule Combined With Glucosamine, notwithstanding the dosage, duration, administration route, administrated approaches or the administration period of therapy.
- Comparator Control
- The comparator was treated with Glucosamine alone.
- Main Outcome
- Improvement in joint pain and function is the main observation item, and which is respectively assessed by the visual analogue scale pain score or a comprehensive evaluation commonly using the Western Ontario and McMaster Universities Arthritis Index scores, Lysholm scores, or Bristol scores. Measures of effect For continuous data will be expressed as the average difference with 95% CI. Weighted mean difference (WMD) will be used for data measured on the same scale and in the same unit; otherwise, standardized mean difference (SMD) will be used.

- The secondary outcome measures include: 1. Efficacy rate, measured as dichotomous outcomes (i.e. no effect, available effect, significant effect, full recovery). 2. Adverse events (AEs) including pain, subcutaneous congestion. Measures of effect For dichotomous data, the risk ratio (RR) with the corresponding 95% CI will be used, while continuous data will be expressed as the average difference with 95% CI. Weighted mean difference (WMD) will be used for data measured on the same scale and in the same unit; otherwise, standardized mean difference (SMD) will be used.
