- Review Question
- Is the treatment of the combination of glucosamine and chondroitin effective for patients with knee osteoarthritis (OA), in terms of pain relief and function recovery? Is the combination of glucosamine and chondroitin a better choice, comparing to NSAIDs?
- Study Type Included
- Only randomized clinical trials (RCTs) will be included.
- Condition Being Studied
- Patients with knee osteoarthritis. Pain relief and function recovery of knee.
- Participant
- Inclusion: Adults with knee osteoarthritis who meet the diagnostic criteria of American College of Rheumatology (ACR) or Chinese Rheumatology Association (CRA). No restrictions for gender, age and severity of knee osteoarthritis. Exclusion: Patients with mental disease or other chronic diseases, or injuries or diseases of knee.

- Intervention
- Inclusion: Studies that investigated the effects of oral administration of the combination of glucosamine and chondroitin among patients with OA, diagnosed through the diagnostic criteria of ACR or CRA. Exclusion: Studies using diagnostic tool other than ACR/CRA diagnostic criteria; Studies in which patients were receiving other interventions like physiotherapy, anti-inflammatory drugs and so on.
- Comparator Control
- Inclusion: Studies that include using one drug alone or Nonsteroidal Antiinflammatory Drugs (NSAIDs), or placebos which are totally same with glucosamine or chondroitin in shape, smell and package. Exclusion: Studies with no control group.
- Main Outcome
- Overall improvement and pain improvement. Measures of effect The overall improvement will be measured by the Western Ontario and McMaster Universities Osteoarthritis Index (WOMAC). Pain will be measured by the Visual Analogue Scale (VAS). The effect sizes will be expressed as Mean Differences (MDs).

- Detail improvement in function, pain and stiffness. The progression of knee degeneration. And the adverse effect. Measures of effect Improvement in function, pain and stiffness will be measured by three subscales of WOMAC, WOMAC function, WOMAC pain and WOMAC stiffness respectively. The progression of knee degeneration will be measured by the radiology manifestation as Joint Space Narrowing (JSN). The effect sizes of all above will be expressed as Mean Differences (MDs). The adverse effect (AE) will be measured by the rate of adverse events. The effect sizes of AE will be expressed as Relative Risks (RR).
