- Title
- Efficacy and Safety of Donepezil, Galantamine, Rivastigmine, Memantine and sodium oligomannate for the Treatment of Alzheimer’s Disease: A Systematic Review and Meta-Analysis
- Review Question
- Is there any difference in safety and effectiveness between anti-dementia drugs and placebo in the treatment of mild to moderate Alzheimer's disease?
- Study Type Included
- Inclusion: (1) double-blind, parallel-group, placebo-controlled, with random assignment to Donepezil, Galantamine, Rivastigmine, Memantine or sodium oligomannate; (2) full-text articles were written by English; (3) participants in the studies were mild to moderate AD; (4) Donepezil, Galantamine, Rivastigmine, Memantine and sodium oligomannatewas compared with placebo directly in trials; (5) participants in the studies were generally over 50 years old Exclusion: (1) participants are not patients with Alzheimer's disease; (2) the drug in studies was not Donepezil, Galantamine, Rivastigmine, Memantine or sodium oligomannate; (3)Non-double-blind, non-randomized, no placebo.
- Condition Being Studied
- Alzheimer’s disease(AD) is the most common cause of dementia, affecting more than 36 million people worldwide. It has been predicted that by 2050, this figure will more than 115 million. This age-associated neurodegenerative disorder has an insidious onset with progressive deterioration in cognition, functional ability, behavior, and mood. Regarding patients with mild-to-moderate AD, the American Psychiatric Association recommends second generation ChEIs, donepezil, galantamine and rivastigmine for mild-to-moderate AD.Memantine and sodium oligomannate were gradually added to the treatment in recent years, but controversy over these drugs still be continued.
- Participant
- Inclusion: (1) participants in the studies were mild-to-moderate AD; (2) Donepezil, Galantamine, Rivastigmine, Memantine and sodium oligomannate were compared with placebo directly in trials; (3) participants in the studies were generally over 50 years old Exclusion: (1) participants are not patients with Alzheimer's disease; (2) the drug in studies was not Donepezil, Galantamine, Rivastigmine, Memantine or sodium oligomannate

- Main Outcome
- We focus on the Alzheimer’s Disease Assessment Scale (ADAS-cog), Alzheimer’s Disease Cooperative Study-Activities of Daily Living (ADCS/ADL), Neuropsychiatric Inventory (NPI), and the Clinician’s Interview Based Assessment of Change-Plus (CIBIC plus) as the primary measure of cognition, function, behavior, and global assessment of change, respectively.
- Outcome Measure
- Additional Outcome
- we recorded the incidence of adverse events (AEs) and serious adverse events (SAEs) as the second outcome to assess the safety of the different drug doses.
