Review Question
Does Letrozole increase ovulation rate, endometrial thickness and the rate of pregnancy compared to Chlomiphene citrate?
Study Type Included
Inclusion criteria: we are planning to include RCT studies in our research work. Exclusion criteria: we are planning to exclude reviews, cases series, cases reports.
Condition Being Studied
Polycystic ovarian syndrome (PCOS) is one of the most common endocrine disorders, characterized by irregular menstrual cycles, hyperandrogenism, and polycystic ovaries. Clomiphene citrate (CC) is the drug of first choice for ovulation induction in women with PCOS in the past 55 years, however, it thins the endometrium via its anti-estrogenic effect and has several side effects (vasomotor reaction, headache, nausea, and breast tenderness) as well. Aromatase inhibitors are new promising ovulation-inducing agents. The efficacy of Letrozole, a selective aromatase inhibitor, is heavily investigated, demonstrating its potential superiority to clomiphene citrate in terms of live birth, pregnancy and ovulation rates, while not having an endometrium thinning effect. Our aim is to compare the efficacy of Letrozole therapy to Clomiphene citrate therapy in women with PCOS.

Primary outcomes: Endometrial thickness, number of dominant follicles, ovulation rates, pregnancy rates Secondary outcomes: Should data volume permit, the following outcomes will be analyzed as well endometrial volume Measures of effect Risk ratios (RR) or Odds ratios (OR) for dichotomous outcomes, Weighted- or standardized mean difference (WMD or SMD) for continuous ones.
