- Title
- Romosozumab, alendronate, risedronate, zoledronic acid, and raloxifene for the treatment of postmenopausal women with severe osteoporosis at high risk of fracture: systematic review and network meta-analysis.
- Review Question
- P - Postmenopausal women with severe osteoporosis at high risk of fracture. I - Romosozumab, alendronate, risedronate, zoledronic acid, and raloxifene. C - Romosozumab, alendronate, risedronate, zoledronic acid, raloxifene or matching placebo. O - mortality; number of patients with at least one new vertebral fracture; number of patients with at least one new non-vertebral fracture; number of patients with discontinuation of treatment due to an adverse event; percentage change in bone mineral density of the femoral neck; percent change in lumbar spine bone mineral density; percent change in total hip bone mineral density; and number of patients with at least one adverse event. S - Randomized Clinical Trials (RCT).
- Study Type Included
- Randomized Clinical Trials (RCT), regardless of phase, duration, or type of analysis (prior or post hoc analysis) will be included. Will be excluded RCT without peer-reviewed and conference poster abstracts.
- Condition Being Studied
- Osteoporosis is a common disease that is characterized by low bone mass with microarchitectural disruption and skeletal fragility, resulting in an increased risk of fracture.
- Participant
- Postmenopausal women diagnosed with severe osteoporosis. According to World Health Organization criteria, osteoporosis is defined as a bone mineral density (BMD) that is 2.5 standard deviations (SD) or more below the mean value for healthy young women (a T-score < -2.5 SD); while severe osteoporosis (or established osteoporosis) is classified as a value of BMD 2.5 SD or more below in the presence of one or more fragility fractures.

- Human Disease Modelled
- Intervention
- Romosozumab, alendronate, risedronate, zoledronic acid, and raloxifene.
- Comparator Control
- Romosozumab, alendronate, risedronate, zoledronic acid, raloxifene, or matching placebo.
- Main Outcome
- Mortality. Number of patients with at least one new vertebral fracture. Number of patients with at least one new non-vertebral fracture. Number of patients with discontinuation of treatment due to an adverse event. Measures of effect Binary outcomes will be summarized as relative risks (RR).
- Outcome Measure
- Additional Outcome
- Percent change in bone mineral density (BMD) of the femoral neck. Percent change BMD of in lumbar spine. Percent change in BMD of total hip. Number of patients with at least one adverse event. Measures of effect Binary outcomes will be summarized as relative risk (RR), while continuous outcomes will be summarized as mean difference.
