Oral etoposide for treatment and/or maintenance treatment of recurrent epithelial ovarian cancer [Cochrane protocol]
Review Question
To assess the clinical therapeutic efficacy and adverse effects of oral etoposide as treatment and/or maintenance treatment in women with recurrent epithelial ovarian cancer.
Study Type Included
We will include published and unpublished randomized controlled trials (RCTs). We will only include non-RCTs that used sensible statistical adjustment in the analysis to reduce the risk of selection bias, especially as our initial search revealed no completed RCT of oral etoposide as treatment of recurrent ovarian cancer. We will include trials irrespective of publication status, year of publication or language.

Participant
We will include women who are 18 years or older with relapsed epithelial ovarian cancer (EOC). This will include women who are platinum-sensitive (disease-free after six months of completing previous chemotherapy), platinum-resistant (relapsed within six months of completing previous chemotherapy) or platinum-refractory (progression on first-line treatment).

Intervention
We will include trials that compare oral etoposide monotherapy, or combination therapy including etoposide, versus another single-agent chemotherapy, combination therapy without oral etoposide, or placebo, used as treatment and/or maintenance treatment in women with recurrent epithelial ovarian cancer. We will exclude trials published only in abstract form or in non‐peer reviewed journals, whereby no further or insufficient information can be procured from the authors. For technical reasons the intervention and comparators fields for Cochrane protocols are identical and each may include information on both interventions and comparators
Comparator Control
We will include trials that compare oral etoposide monotherapy, or combination therapy including etoposide, versus another single-agent chemotherapy, combination therapy without oral etoposide, or placebo, used as treatment and/or maintenance treatment in women with recurrent epithelial ovarian cancer. Main Outcome
<ol> Overall survival (OS), defined as death from all causes, which will be assessed from the time women are randomized in the study Progression-free survival (PFS), defined as survival until disease progression, which will be assessed from the time of randomization until the progression or recurrence of the disease or death</ol>

<ol> Objective response rate (ORR) Quality of life (QoL), measured using a scale that has been validated for use in cancer patients treated with anti-cancer drugs (<link ref=REF-Aaronson-1993 type=REFERENCE/>Aaronson 1993; <link ref=REF-Cella-1993 type=REFERENCE/>Cella 1993; <link ref=REF-Schipper-1984 type=REFERENCE/>Schipper 1984) Serious adverse events (AEs), classified according to Common Terminology Criteria for Adverse Events (CTCAE) version 3.0 (<link ref=REF-CTCAE-2006 type=REFERENCE/>CTCAE 2006). Grades of toxicity will be extracted and grouped as:<ol> haematological (leukopenia, anaemia, thrombocytopenia, haemorrhage); cardiac (bradycardia, atrial fibrillation); gastrointestinal (nausea, vomiting, anorexia, diarrhoea, liver, proctitis); genitourinary; skin (stomatitis, mucositis, alopecia, allergy); neurological (peripheral and central); pulmonary; incidence of secondary malignancy.</ol></ol>
