- Title
- Comparison of high-dose dual therapy,bismuth-based quadruple therapy and non-bismuth quadruple therapy for Helicobacter pylori infection:a network meta-analysis of randomized controlled trials
- Review Question
- To study the efficacy and safety of Helicobacter pylori eradication therapies including high-dose dual therapy, bismuth-based quadruple therapy and non-bismuth quadruple therapy
- Study Type Included
- Randomized controlled trials to assess the efficacy and safety of the treatments for Hp infection were included.
- Condition Being Studied
- Helicobacter pylori infection. First-line treatment for adults.
- Participant
- Inclusion: Participants were considered eligible if they met following criteria: 1. adults from 18 to 75 years old; 2.H. pylori infection diagnosed through at least one standard methods (urea breath test, bacterial culture, rapid urea test, histological examination);3.treatment-naive patients for H.pylori . Exclusion:These participants were excluded:1.allergy to antibiotics such as Amoxicillin, Clarithromycin, Metronidazole, tetracycline, furazolidone;2.use of adrenocorticosteroids, nonsteroidal anti-inflammatory drugs, and anticoagulants; 3.patients with other complications such as liver disease, cardiac vascular disease, lung disease, kidney disease, metabolic disease, mental illness, or malignant tumors, that might interfere with the evaluation of study treatment; 4. pregnant and breastfeeding women ;5. participation in other clinical studies within 3 months;6. incomplete follow-up or without informed consents before enrollment of the study.
- Animal
- Human Disease Modelled
- Intervention
- 1.High-dose dual therapy including PPI and one antibiotic, increasing both dosage and frequency of administration; 2.bismuth-based quadruple therapy (PPI + bismuth + two antibiotics with various combinations) 3.non-bismuth quadruple therapy was divided into sequential therapy (PPI + antibiotic for the first 5 or 7 days, then 5 or 7 days PPI and another two antibiotics); concomitant therapy (PPI combined three antibiotics such as amoxicillin, clarithromycin, and a nitroimidazole administered concomitantly for 10 or 14 days), and hybrid therapy (the first 5 or 7 day the same as sequential therapy, followed by 5 or 7 days the same as concomitant therapy). H2-blocker containing therapy and use of probiotics, herbs were excluded.
- Comparator Control
- Specified above.
- Main Outcome
- The eradication rate of Hp by ITT and PP analysis Measures of effect at least four weeks after completion of regimens, determined by the UBT assay or other methods according to the studies.
- Outcome Measure
- Additional Outcome
- Adverse events and adherence. Measures of effect Adverse events were recorded by participants as instructed according to the influence on their daily activities. adherence was measured by a patient pill counts, ≥80% was considered good compliance, and <80% was defined as poor compliance.
