性状
本品呈椭圆形或近卵形,长2~3cm,直径1.4~2cm。表面黑褐色或棕褐色,稍有光泽,具不规则的皱褶,基部有果梗痕。果肉薄,棕褐色,质硬而脆。核近卵形,黄棕色,顶端有5个(偶有4个或6个)明显的小孔,每孔内各含种子1枚。气微,味酸。
检查
水分 取本品去核粉末,照水分测定法(通则0832第二法)测定,不得过13.0%。 总灰分 取本品去核粉末,照灰分测定法(通则2302)测定,不得过6.5%。
贮藏
置阴凉干燥处。
鉴别
(1)本品粉末棕色。内果皮石细胞呈类圆形、椭圆形、梭形、长方形或不规则形,有的延长呈纤维状或有分枝,直径14~72μm,长25~294μm,壁厚,孔沟明显,胞腔内含淡黄棕色或黄褐色物。内果皮纤维木化,多上下层纵横交错排列,壁厚或稍厚,有的胞腔内含黄棕色物。外果皮细胞表面观呈类多角形,胞腔内含棕色物;断面观细胞呈类长方形,径向延长,外壁及径向壁角质化增厚。中果皮薄壁细胞含草酸钙簇晶和少数方晶,簇晶直径17~42μm,方晶菱形或不规则形,长10~48μm,直径7~27μm。 (2)取本品粉末2g,加70%乙醇20ml,加热回流15分钟,滤过,滤液蒸干,加乙酸乙酯10ml使溶解,滤过,取滤液1ml,置蒸发皿中,蒸干,加硼酸饱和的丙酮溶液与10%枸橼酸丙酮溶液各1ml,显黄绿色,继续蒸干,置紫外光灯(365nm)下观察,显黄绿色荧光;另取滤液1ml,置试管中,蒸干,加甲醇1ml使溶解,加三氯化铝试液3~4滴,溶液黄色略加深,点于滤纸上,置紫外光灯(365nm)下观察,显黄绿色荧光。 (3)取本品粉末5g,加70%乙醇30ml,加热回流30分钟,滤过,滤液蒸至约2ml,加水5ml使溶解,用乙醚振摇提取2次,每次15ml,合并乙醚液,蒸干,残渣加乙酸乙酯1ml使溶解,作为供试品溶液。另取没食子酸对照品,加无水乙醇制成每1ml含1mg的溶液,作为对照品溶液。照薄层色谱法(通则0502)试验,吸取供试品溶液10μl、对照品溶液5μl,分别点于同一硅胶G薄层板上,以三氯甲烷-丙酮-甲酸(7∶2∶1)为展开剂,展开,展距15cm,取出,晾干,置氨蒸气中熏至斑点显色清晰。供试品色谱中,在与对照品色谱相应的位置上,显相同颜色的斑点。
浸出物
取本品去核粉末,照醇溶性浸出物测定法(通则2201)项下的热浸法测定,用稀乙醇作溶剂,不得少于28.0%。
含量测定
照高效液相色谱法(通则0512)测定。 色谱条件与系统适用性试验 以十八烷基硅烷键合硅胶为填充剂;以甲醇-水-冰醋酸(1∶99∶0.3)为流动相;检测波长为270nm;柱温为30℃。理论板数按没食子酸峰计算应不低于3000。 对照品溶液的制备 取没食子酸对照品适量,精密称定,加甲醇制成每1ml含60μg的溶液,即得。 供试品溶液的制备 取本品去核粉末(过二号筛)约1g,精密称定,置具塞锥形瓶中,精密加入70%甲醇20ml,称定重量,加热回流1小时,放冷,再称定重量,用70%甲醇补足减失的重量,摇匀,滤过,取续滤液,即得。 测定法 分别精密吸取对照品溶液与供试品溶液各10μl,注入液相色谱仪,测定,即得。 本品去核后按干燥品计算,含没食子酸(C7H6O5)不得少于0.060%。
功能与主治
行气活血,养心,安神。用于气滞血瘀,胸痹作痛,心悸气短,心神不安。
