本品呈不规则的段。茎短圆柱形，表面绿褐色，有数条纵棱，被丝状毛；切面灰白色，髓部疏松或中空。叶皱缩，多破碎，边缘具不等长的针刺；两面均具灰白色丝状毛。头状花序多破碎。气微，味淡。

检查

杂质不得过2%（通则2301）。水分不得过13.0%（通则0832第二法）。酸不溶性灰分不得过3.0%（通则2302）。

炮制

除去杂质，抢水洗或润软后，切段，干燥。

贮藏

置通风干燥处。

鉴别

（1）叶表面观：上表皮细胞多角形；下表皮细胞类长方形，垂周壁波状弯曲。气孔不定式或不等式，副卫细胞3～5个。非腺毛4～18细胞，顶端细胞细长而扭曲，直径约7μm，壁具交错的角质纹理。（2）取本品粉末1g，加甲醇10ml，超声处理30分钟，滤过，滤液蒸干，残渣加甲醇2ml使溶解，作为供试品溶液。另取大蓟对照药材1g，同法制成对照药材溶液。照薄层色谱法（通则0502）试验，吸取上述两种溶液各1～2μl，分别点于同一聚酰胺薄膜上，以乙酰丙酮-丁酮-乙醇-水（1∶3∶3∶13）为展开剂，展开，取出，晾干，喷以三氯化铝试液，晾干，置紫外光灯（365nm）下检视。供试品色谱中，在与对照药材色谱相应的位置上，显相同颜色的荧光主斑点。

本品为不规则长圆形或圆形厚片。外表皮棕褐色，粗糙，有皱纹。切面棕黄色或棕褐色，纤维性。质坚硬。微有醋气，味微苦涩。

检查

水分不得过11.0%（通则0832第二法）。

注意

孕妇禁用；不宜与甘草同用。

炮制

京大戟除去杂质，洗净，润透，切厚片，干燥。

贮藏

置干燥处，防蛀。

鉴别

（1）本品粉末淡黄色。淀粉粒单粒类圆形或卵圆形，直径3～15μm，脐点点状或裂缝状；复粒由2～3分粒组成。草酸钙簇晶直径19～40μm。具缘纹孔导管和网纹导管较多见，直径26～50μm。纤维单个或成束，壁较厚，非木化。无节乳管多碎断，内含黄色微细颗粒状乳汁。（2）取本品手切薄片2片，一片加冰醋酸与硫酸各1滴，置显微镜下观察，在韧皮部乳管群处呈现红色，5分钟后渐褪去；另一片加氢氧化钾试液，呈棕黄色。（3）取本品粉末0.5g，加石油醚（60～90℃）5ml，浸渍1小时，滤过，滤液浓缩至1ml，作为供试品溶液。另取京大戟对照药材1g，同法制成对照药材溶液。再取大戟二烯醇对照品，加甲醇制成每1ml含1mg的溶液，作为对照品溶液。照薄层色谱法（通则0502）试验，吸取上述三种溶液各2μl，分别点于同一硅胶G薄层板上，以石油醚（60～90℃）-丙酮（7∶1）为展开剂，展开，取出，晾干，喷以10%硫酸乙醇溶液，在105℃加热至斑点显色清晰，分别置日光和紫外光灯（365nm）下检视。供试品色谱中，在与对照药材色谱和对照品色谱相应的位置上，显相同颜色的斑点或荧光斑点。

浸出物

照醇溶性浸出物测定法（通则2201）项下的冷浸法测定，用乙醇作溶剂，不得少于8.0%。

含量测定

照高效液相色谱法（通则0512）测定。色谱条件与系统适用性试验以辛烷基硅烷键合硅胶为填充剂；以乙腈-水（92∶8）为流动相；检测波长为210nm。理论板数按大戟二烯醇峰计算应不低于5000。对照品溶液的制备取大戟二烯醇对照品适量，精密称定，加甲醇制成每1ml含0.2mg的溶液，即得。供试品溶液的制备取本品粉末（过四号筛）约1g，精密称定，置具塞锥形瓶中，精密加入乙醇50ml，密塞，称定重量，超声处理（功率200W，频率40kHz）30分钟，放冷，再称定重量，用乙醇补足减失的重量，摇匀，滤过，精密量取续滤液10ml，蒸干，残渣加甲醇溶解，转移至5ml量瓶中，加甲醇稀释至刻度，摇匀，滤过，取续滤液，即得。测定法分别精密吸取对照品溶液10μl与供试品溶液5～10μl，注入液相色谱仪，测定，即得。本品按干燥品计算，含大戟二烯醇（C30H50O）不得少于0.60%。饮片
